Development and intercomparison of single and multicompartment physiologically-based toxicokinetic models: Implications for model selection and tiered modeling frameworks

Summary: This study presents the development and comparison of generic PBTK models for humans, which showed consistent results for key toxicokinetic parameters between one-compartment and multi-compartment models, but discrepancies may arise in cases of rapid biotransformation. Differences are most pronounced for neutral organics and least for acidic IOCs.

A multispecies toxicokinetic model based on data from 69 freshwater fish species in Canada was developed to predict bioconcentration, showing promising results with some tendency of overestimating measured values. This bottom-up approach potentially provides more accurate predictions for chemical risk assessments.
