A Gene Expression Biomarker Identifies Chemical Modulators of Estrogen Receptor α in an MCF-7 Microarray Compendium

John Rooney et al.

Summary: Identification of potential ER alpha modulators using a 46 gene biomarker, screening a large microarray compendium and the Connectivity Map 2.0 collection, and further examination through various assays revealed known and novel activators and suppressors of ER alpha in MCF-7 cells. This strategy will aid in identifying environmentally relevant ER alpha modulators in future high-throughput transcriptomic screens.

Deriving predicted no-effect concentrations (PNECs) using a novel assessment factor method

Alexander Okonski et al.

Summary: The traditional assessment factor method has been criticized for its uncertainty, inaccuracy, and lack of transparency, prompting the proposal of a novel method. This new method breaks the factor down into three components and can include read-across and modeled data, better managing atypical datasets and endpoints, while also improving consistency among risk assessors and increasing transparency for stakeholders.

Towards a comprehensive characterisation of the human internal chemical exposome: Challenges and perspectives

Arthur David et al.

Summary: The use of high-resolution mass spectrometry to characterize the human internal chemical exposome provides unprecedented precision in investigating the environmental etiology of chronic diseases. However, improvements are needed in the flexibility, sensitivity, and automation of analytical techniques. Additionally, challenges exist in assessing the dynamic nature of the internal chemical exposome, establishing international collaboration, high level networking, and sustainable research infrastructure.

High-Throughput Transcriptomics Platform for Screening Environmental Chemicals

Joshua A. Harrill et al.

Summary: This study evaluated the TempO-Seq assay for HTTr concentration-response screening of chemicals in the human-derived MCF7 cell model. A robust bioinformatics pipeline was developed using open-source tools, along with a novel gene expression signature-based concentration-response modeling approach. Analysis showed highly reproducible differential gene expression signatures, along with in vitro transcriptional biological pathway altering concentrations closely aligned with previous high-throughput screening assays.

Identification of non-validated endocrine disrupting chemical characterization methods by screening of the literature using artificial intelligence and by database exploration

Elias Zgheib et al.

Summary: Exposure to endocrine disrupting chemicals (EDCs) poses a significant public health threat, with a range of adverse health outcomes associated. The current regulatory tests do not cover all endocrine pathways, necessitating the search for more comprehensive testing methods for regulatory pre-validation. Through a combination of automated and manual screening, several non-validated experimental methods were identified, covering multiple endocrine pathways.
