Summary: This study characterized a group of bacteria associated with sponges through metagenomics, phylogenetic analyses, metatranscriptomics, and fluorescence in situ hybridization microscopy. The newly identified gammaproteobacterial order (U)Tethybacterales shows a deep branching within Gammaproteobacteria and almost exclusive presence in sponges, indicating early symbiosis and subsequent functional radiation. The diverse lifestyles and morphologies of (U)Tethybacterales species reflect their varied substrate preferences and localization within sponge tissues.
