Electrochemiluminescence Microscopy of Cells: Essential Role of Surface Regeneration

Dongni Han et al.

Summary: Electrochemiluminescence (ECL) microscopy is successfully used for imaging cells, micro-/nano-objects, and electrochemical processes. The decrease of ECL signal during successive image recordings is a key limitation for ECL microscopy development. An electrochemical approach involving cathodic regenerative treatment can restore the ECL signal intensity, allowing for continuous image recording without signal loss.

Electrochemiluminescence Loss in Photobleaching

Dongni Han et al.

Summary: The study investigated the effects of photobleaching on electrochemiluminescence (ECL) for the first time, demonstrating a linear correlation between ECL decrease and photoluminescence (PL) loss. The presented strategy provides valuable information on ECL excited state fundamentals and opens new opportunities for exploring cellular membranes by combining ECL microscopy with photobleaching techniques such as FRAP or FLIP methods.

Bio-Coreactant-Enhanced Electrochemiluminescence Microscopy of Intracellular Structure and Transport

Cheng Ma et al.

Summary: The bio-coreactant-enhanced electrochemiluminescence (ECL) microscopy allows for ECL imaging of intracellular structure and dynamic transport by using Ru(bpy)(3)(2+) and intracellular biomolecules. The research reveals the universal edge effect of cellular electroporation, as well as the dynamic transport of Ru(bpy)(3)(3+) in different cellular compartments and its correlation with the actin cytoskeleton.

Spatially Selective Imaging of Cell-Matrix and Cell-Cell Junctions by Electrochemiluminescence

Hao Ding et al.

Summary: A novel method for spatially selective imaging of cell junctions by regulating the thickness of the electrochemiluminescence (ECL) layer through control of luminophore and co-reactant concentrations has been reported. Low luminophore concentration reveals cell-matrix adhesions, while high concentration allows sequential imaging of cell-matrix and cell-cell junctions. This strategy not only provides new insights into ECL mechanisms but also expands the potential applications of ECL microscopy in bioimaging.

Electrocatalyst Screening on a Massive Array of Closed Bipolar Microelectrodes

Todd J. Anderson et al.

Summary: In this study, a novel electroanalytical platform utilizing bipolar ultramicroelectrode arrays and electrochemiluminescence imaging was developed for rapid screening of electrocatalysts. By electrodeposition of a radial gradient of catalytic metal on carbon-gold bipolar UME arrays, the impact of varying Ni(OH)2 coverage on the catalytic activity for the hydrogen evolution reaction (HER) was investigated. Future work will focus on expanding this platform to screen various metal alloys by incorporating multiple metal salts delivery during the gradient electrodeposition process.

Shadow Electrochemiluminescence Microscopy of Single Mitochondria

Yumeng Ma et al.

Summary: This study introduces a label-free shadow electrochemiluminescence (ECL) microscopy technique to image single living mitochondria deposited on electrode surface. By utilizing negative optical contrast and local diffusional hindrance, this approach visualizes individual mitochondria and validates their functionality with classical fluorescent biomarkers. The versatility and extreme sensitivity of this method make it promising for imaging subcellular structures while alleviating photobleaching and phototoxicity issues associated with conventional microscopy.

Elena Villani et al.

Summary: In this study, electrochemiluminescence (ECL) was successfully generated from entirely pure conducting polymer films in alkaline aqueous solution, using PEDOT and PPy films prepared by constant current electrolysis as bipolar electrodes. ECL intensity profiles showed gradient emission on both materials, with different light-emitting regions. Imaging analysis allowed evaluation of ECL behavior of both CPs as a function of applied voltage and conductivity, showing potential applications in ECL and bipolar electrochemistry fields.

3D in vitro culture system to study collective migration in mammary organoid epithelium

Yunzhe Lu et al.

Summary: A new in vitro culture system for mammary gland organoid directional migration in response to FGF10 concentration gradient has been established, with a step-by-step protocol provided for preparing organoids and setting up the 3D culture system. The main technical difficulties in conducting the 3D migration assay involve selecting appropriate sizes of epithelial organoids and manually pairing organoids and FGF10-soaked beads within a desired distance.
