The Second-Generation Click-to-Sense Probe for Intraoperative Diagnosis of Breast Cancer Tissues Based on Acrolein Targeting

Summary: Recently, it has been discovered that a bulky 2,6-diisopropylphenyl azide shows higher reactivity towards acrolein compared to a simple phenyl azide. In this study, tetramethylrhodamine (TAMRA) was conjugated to the bulky phenyl azide to create a second-generation click-to-sense (CTS) probe for analyzing cancer by targeting cellular acrolein. The results showed that this second-generation CTS probe could distinguish breast cancer tissues more sensitively than the first-generation CTS probe. Additionally, the addition of Rh-2(OAc)(4) to the system promoted the formation of a metal carbenoid intermediate, which accelerated the covalent attachment of the probe to an organelle in cancer cells.
