Radio Signals from Live Cells: The Coming of Age of In-Cell Solution NMR

Summary: A detailed understanding of the complex processes that contribute to the functionality of cells and organisms is crucial for disease comprehension and drug development. Among the available structural techniques, solution NMR is the only method capable of investigating macromolecular structures and dynamic behaviors directly within living cells. Over the past 20 years, in-cell NMR has emerged as a powerful tool, allowing researchers to study cell samples, isotopic labeling, and bioreactor devices, thus enhancing the lifetime of cells for real-time monitoring of intracellular metabolites and proteins.

In-Cell NMR Spectroscopy of Functional Riboswitch Aptamers in Eukaryotic Cells

Summary: This study reports the in-cell NMR-spectroscopic observation of the binding of the bacterial riboswitch ligand with the aptamer domain in eukaryotic cells, demonstrating the maintenance of binding mode in eukaryotic cellular environment. Additionally, the study shows that the in-cell NMR setup can be used for characterization of large, non-modified RNA molecules in cells.

Through-Space Scalar 19F-19F Couplings between Fluorinated Noncanonical Amino Acids for the Detection of Specific Contacts in Proteins

Summary: In this study, it was demonstrated that fluorine atoms in noncanonical amino acids positioned in the hydrophobic core or on the surface of a protein can be linked by scalar through-space F-19-F-19 couplings, even if the F-19 spins are separated by more than the van der Waals distance. The use of fluorinated amino acids and F-19-F-19 interactions offer a straightforward way to probe the spatial proximity of selected sites in proteins, without requiring H-1 NMR resonance assignments.

Protein in-cell NMR spectroscopy at 1.2 GHz

Summary: In-cell NMR spectroscopy provides valuable information on biological macromolecules at atomic resolution in their native cellular environment, but the low sensitivity of NMR limits its applicability. The recently developed commercial 1.2 GHz NMR spectrometer is expected to bring significant benefits, although cell samples may suffer detrimental effects at ultrahigh fields.

Determination of intracellular protein-ligand binding affinity by competition binding in-cell NMR

Summary: Structure-based drug development faces challenges in cellular and animal models due to various factors such as low cell penetrance, off-target binding, and conformational changes of the target protein. In this study, in-cell NMR spectroscopy was used to measure intracellular dissociation constants, showing that the method can effectively determine the binding affinity of compounds to intracellular targets.

Towards Profiling of the G-Quadruplex Targeting Drugs in the Living Human Cells Using NMR Spectroscopy

Summary: H-1 and F-19-detected in-cell NMR spectroscopy is used to assess the application potential of drugs/ligands targeting DNA G-quadruplexes. Despite challenges such as signal broadening and overlap in in-cell NMR spectra, this method can identify drugs that lose their ability to bind G-quadruplexes in the native environment. The adjustment of the in-cell NMR approach to a 3,5-bis(trifluoromethyl)phenyl probe for F-19 detection appears to be a promising strategy for profiling G-quadruplex-ligand interactions in living cells.
