Determining the effect of pretreatments on freeze resistance and survival of cryopreserved temperate fruit tree dormant buds

Summary: Freeze resistance is crucial for successful dormant bud cryopreservation, and different pretreatments such as sucrose exposure and desiccation have been shown to improve postcryogenic viability in various tree fruit species. Desiccation to 30% moisture content was found to have the greatest impact on increasing freeze resistance and cryosurvival across most fruit species tested. Gradual reduction of moisture content levels showed increased freeze resistance in peach, although severe bud cracking was observed, indicating the complexity of cryorecovery outcomes.
