Qiu-shi Xie et al.

Summary: Short-chain fatty acids (SCFAs) have opposite effects on the expression and function of intestinal P-gp and BCRP; butyrate downregulates P-gp expression and function possibly via inhibiting HDAC/NF-kappa B pathways; butyrate induces BCRP expression and function partly via PPAR gamma activation.

Pre-incubation with OATP1B1 and OATP1B3 inhibitors potentiates inhibitory effects in physiologically relevant sandwich-cultured primary human hepatocytes

Taleah Farasyn et al.

Summary: This study demonstrates that pre-incubation with OATP1B inhibitors potentiates inhibitory effects in physiologically relevant primary human hepatocytes, supporting the rationale of the current US FDA draft guidance. IC50 values after inhibitor-preincubation in transporter-expressing cell lines may be used for DDI prediction for the purpose of mitigating false-negative OATP-mediated DDI prediction.

Regulation of OATP1B1 Function by Tyrosine Kinase-mediated Phosphorylation

Elizabeth R. Hayden et al.

Summary: OATP1B1 is a major drug uptake transporter in the liver, and its activity can be significantly inhibited by various TKIs. The inhibition of OATP1B1 by TKIs can lead to reduced phosphorylation levels, affecting its transport function and pharmacokinetics of substrates like rosuvastatin. Identified Src kinase LYN as a potential regulator of OATP1B1 activity, highly sensitive to inhibition by clinically relevant concentrations of TKIs.

Molecular Regulation of Canalicular ABC Transporters

Amel Ben Saad et al.

Summary: ABC transporters expressed at the canalicular membrane of hepatocytes are crucial for bile secretion by secreting bile acids, phosphatidylcholine, cholesterol, drugs, and other compounds. Dysfunctions in these transporters lead to rare, severe biliary diseases, emphasizing the importance of studying their biology for developing new therapies.

Summary: The bile salt export pump (BSEP/ABCB11) plays a crucial role in bile salt secretion by controlling the efflux of bile salts from hepatocytes into the bile. Research efforts have been focused on understanding the physiological and pathological functions and regulatory mechanisms of BSEP. Several liver diseases have been linked to BSEP expression and dysfunction.

RNA editing enzyme ADAR1 governs the circadian expression of P-glycoprotein in human renal cells by regulating alternative splicing of the ABCB1 gene

Yuji Omata et al.

Summary: The study revealed that ADAR1-mediated regulation of alternative splicing of the ABCB1 gene is a key mechanism for the circadian expression of P-glycoprotein (P-gp) in human renal proximal tubular epithelial cells (RPTECs). This regulatory mechanism may explain the dosing time-dependent variations in the renal elimination of P-gp substrates.

Protein kinase C regulates organic anion transporter 1 through phosphorylating ubiquitin ligase Nedd4-2

Zhou Yu et al.

Summary: This study investigates the role of Nedd4-2 phosphorylation in PKC regulation of OAT1. The results indicate that PKC activation enhances Nedd4-2 phosphorylation, leading to increased OAT1 ubiquitination and decreased cell surface expression and transport function. The PKC effects can be reversed by a PKC-specific inhibitor. Additionally, it was found that the quadruple mutant of Nedd4-2 partially blocks PKC-induced phosphorylation and the subsequent effects on OAT1 transport activity.
