The long-circulating effect of pegylated nanoparticles revisited via simultaneous monitoring of both the drug payloads and nanocarriers

Summary: The study revisits the long-circulating effect of mPEG-PCL nanoparticles by monitoring the movement of drug payloads and nanocarriers. It demonstrates that pegylation can extend the circulation time of nanocarriers in the blood, although nanocarriers with dense PEG decoration are cleared quickly, and DOX is cleared faster than the nanocarriers. The study also shows that hepatic accumulation is the highest among all organs investigated but is inversely proportional to blood circulation time, suggesting possible leakage of DOX from the nanocarriers. Reduction in particle size and pegylation prove to be effective in extending circulation time and reducing uptake by organs of the mononuclear phagocytic system.
