Fluorescence-activated droplet sorting of PET degrading microorganisms

Yuxin Qiao et al.

Summary: A high-throughput screening pipeline was developed to identify PET-degrading microorganisms or enzymes, successfully isolating species capable of degrading PET from environmental samples and genetically deriving and validating two putative PETases. The FADS pipeline shows promise for discovering new plastic-degrading microorganisms and enzymes, potentially aiding in the directed evolution of degrading enzymes through synthetic biology.

A Titratable Cell Lysis-on-Demand System for Droplet-Compartmentalized Ultrahigh-Throughput Screening in Functional Metagenomics and Directed Evolution

Che Fai Alex Wong et al.

Summary: Water-in-oil emulsion droplets are a suitable format for high-throughput screening, allowing characterization of libraries with over 10^7 members in a day. The system compartmentalizes single library members in droplets, generated by microfluidic devices, for testing of target biocatalysts. Cell lysis in Escherichia coli can be controlled by adjusting externally added inducer concentrations, with the ability to evaluate the process at both population and single-cell levels.
