Synthesis of NNN Chiral Ruthenium Complexes and Their Cytotoxicity Studies

Pran Gobinda Nandi et al.

Summary: The synthesis and characterization of chiral pincer-ruthenium complexes of ((NNN)-N-R2)RuCl2 (PPh3) (R = 3-methylbutyl and 3,3dimethylbutyl) were reported. Cytotoxicity studies showed these complexes inhibited normal and cancer cell growth in a dose-dependent manner. Various assays revealed that the treatment with pincer-ruthenium complexes induced a redox imbalance in cells by upregulating ROS generation and causing necrotic cell death.

Ruthenium(II) Diphosphine Complexes with Mercapto Ligands That Inhibit Topoisomerase IB and Suppress Tumor Growth In Vivo

Monize M. da Silva et al.

Summary: Ruthenium(II) complexes with unique ligands showed high cytotoxic activity against specific cancer cell lines, while demonstrating selectivity towards non-tumor cells. The complexes inhibited human topoisomerase IB through a multitarget mechanism, with one complex effectively targeting cleavage and religation steps. Molecular docking revealed interactions with active sites on enzymes and DNA. Complex Ru3 displayed significant antitumor activity against melanoma in mouse models without causing DNA damage.

Intercalation Ability of Novel Monofunctional Platinum Anticancer Drugs: A Key Step in Their Biological Action

Daniele Veclani et al.

Summary: Phenanthriplatin is a monovalent platinum(II)-based complex with high cytotoxicity against cancer cells, its mechanism of action involves fast intercalation into DNA bases to form a covalent bond at a guanine site, leading to permanent DNA damage. The impact of DNA sequence and backbone structure on drug anchoring into the double helix was also discussed.
