Cross-validation strategies in QSPR modelling of chemical reactions

Summary: This article discusses the issue of optimistic bias in the conventional k-fold cross-validation procedure for quantitative structure-property relationship models for reactions, and proposes two new strategies, 'transformation-out' CV and 'solvent-out' CV, to provide unbiased estimation of predictive performance for novel types of structural transformations in chemical reactions. These strategies have been successfully applied to predict rate constants of various reactions and are implemented in the open-source software package CIMtools.
