Modeling, optimization, and comparable efficacy of T cell and hematopoietic stem cell gene editing for treating hyper-IgM syndrome

Valentina Vavassori et al.

Summary: The study developed a comprehensive editing strategy to correct the CD40LG gene in T cells and HSPC, demonstrating the therapeutic potential for treating HIGM1. Results showed that edited patient-derived CD4 T cells and adoptive transfer of wild-type T cells effectively restored immune functions and protected mice from disease.

Gene therapy using haematopoietic stem and progenitor cells

Giuliana Ferrari et al.

Summary: Haematopoietic stem and progenitor cell (HSPC) gene therapy has shown to be effective in treating monogenic disorders of the blood system, including primary immunodeficiencies and beta-thalassaemia. Additionally, site-specific genome modification of HSPCs using gene editing techniques has great clinical promise.

Single-dose MGTA-145/plerixafor leads to efficient mobilization and in vivo transduction of HSCs with thalassemia correction in mice

Chang Li et al.

Summary: A new in vivo hemopoietic stem cell gene therapy approach has been developed without the need for myelosuppressive conditioning and autologous HSC transplantation, involving HSC mobilization and a helper-dependent adenovirus vector system. The mobilization regimen using truncated GRO-beta and plerixafor resulted in robust HSC mobilization with less toxicity. In a mouse model, in vivo HSC transduction after mobilization led to complete correction of thalassemia, demonstrating the potential for long-term HSC repopulation.

Daniele Canarutto et al.

Summary: The study presents safety and efficacy data of PBSC collection in 45 pediatric patients for ex vivo gene transfer. The results show that PBSC collection compares well to BM harvest in terms of CD34(+) cell yields for the purpose of gene therapy, with a favorable safety profile.

Gene Editing of Hematopoietic Stem Cells: Hopes and Hurdles Toward Clinical Translation

Samuele Ferrari et al.

Summary: Gene therapies based on integrating vectors have shown outstanding results in the field of hematology for treating various human diseases. With the introduction of novel programmable nucleases like CRISPR/Cas9, gene therapy has moved beyond semi-random gene addition to precise modification of the genome, promising safer manipulation of genes. Research on ex vivo gene editing with programmable nucleases in human hematopoietic stem and progenitor cells (HSPCs) highlights the potential benefits and current obstacles towards clinically translating gene editing for hematological diseases.
