Fascin promotes lung cancer growth and metastasis by enhancing glycolysis and PFKFB3 expression

Shengchen Lin et al.

Summary: Fascin, a pro-metastatic actin-bundling protein, promotes cancer cell migration and invasion by increasing glycolysis in lung cancer through activation of yes-associated protein 1 (YAP1). The fascin-YAP1-PFKFB3 axis is likely conserved across different types of cancers, and pharmacological inhibitors of fascin may be used to reprogram cancer metabolism in lung and potentially other cancers with fascin upregulation.

Inhibition of PFKFB3 induces cell death and synergistically enhances chemosensitivity in endometrial cancer

Yinan Xiao et al.

Summary: The inhibition of PFKFB3 with PFK158 can reduce chemoresistance in endometrial cancer cell lines by lowering cell proliferation, increasing sensitivity to chemotherapy, inhibiting glucose metabolism, and promoting apoptosis and autophagy-induced cell death. The combination of PFK158 with chemotherapy drugs can enhance chemosensitivity by affecting cell signaling pathways, ultimately leading to suppressed tumor growth.

Inhibition of glycolytic activator PFKFB3 suppresses tumor growth and induces tumor vessel normalization in hepatocellular carcinoma

Kenichi Matsumoto et al.

Summary: The study found that high expression of PFKFB3 in both tumor cells and tumor endothelial cells is significantly associated with poor prognosis in hepatocellular carcinoma (HCC). PFK15 was shown to suppress proliferation of HCC cells and tumor endothelial cells, induce apoptosis, and promote tumor vessel normalization, indicating a promising strategy for inhibiting tumor growth.

Role of PFKFB3 and PFKFB4 in Cancer: Genetic Basis, Impact on Disease Development/Progression, and Potential as Therapeutic Targets

Krzysztof Kotowski et al.

Summary: Glycolysis is a crucial metabolic process in rapidly proliferating cells, such as cancer cells, with PFK-1 being a key rate-limiting enzyme regulated primarily by F-2,6-BP. PFK-2/FBPase-2 is a bifunctional enzyme responsible for F-2,6-BP synthesis and degradation, and alterations in PFKFB isozymes play important roles in various diseases, particularly in cancer.

Effects of the Novel PFKFB3 Inhibitor KAN0438757 on Colorectal Cancer Cells and Its Systemic Toxicity Evaluation In Vivo

Tiago De Oliveira et al.

Summary: The PFKFB3 inhibitor KAN0438757 significantly reduced CRC cell migration, invasion and survival, and showed significant effects on growth in patient-derived cancer organoids, while sparing normal colon organoids and healthy mice from high toxicity. These findings support further translational studies to evaluate KAN0438757 in CRC therapy.

Inhibition of Glycolysis Suppresses Cell Proliferation and Tumor Progression In Vivo: Perspectives for Chronotherapy

Jana Horvathova et al.

Summary: The inhibition of glycolysis by targeting PFKFB3 with PFK15 shows promising results in suppressing cell proliferation and inducing apoptosis in tumor and endothelial cells. Both in vitro experiments and animal studies demonstrate a reduction in tumor size with PFK15 treatment, with its effectiveness being circadian-dependent.
