Genome-Wide Identification and Expression Profiling of the PDI Gene Family Reveals Their Probable Involvement in Abiotic Stress Tolerance in Tomato (Solanum lycopersicum L.)

Antt Htet Wai et al.

Summary: Protein disulfide isomerases (PDI) and PDI-like proteins play a crucial role in catalyzing protein disulfide bond formation and isomerization in the endoplasmic reticulum, preventing the accumulation of misfolded proteins under abiotic stress conditions. In tomato, 19 PDI genes were identified, with most showing differential expression across organs and developmental stages, and being highly induced by heat, salt, and ABA treatments. The predominant expression of specific PDI genes in response to abiotic stress and ABA treatment suggests their regulatory roles in abiotic stress tolerance in a tomato.

MBD2 serves as a viable target against pulmonary fibrosis by inhibiting macrophage M2 program

Yi Wang et al.

Summary: Despite extensive studies, the mechanisms underlying PF remain poorly understood. This study found altered MBD2 expression in macrophages in lungs from different types of PF patients and mice with PF, and Mbd2 deficiency protected against BLM-induced PF. Mbd2 selectively influenced TGF-β1 production and M2 macrophage program through Ship promoter binding in macrophages.

Overexpression of a methyl-CpG-binding protein gene OsMBD707 leads to larger tiller angles and reduced photoperiod sensitivity in rice

Mengyu Qu et al.

Summary: In this study, the expression patterns of OsMBD family genes in rice were explored, and 13 OsMBDs with active expression in various rice tissues were identified. The function of a rice class I MBD protein OsMBD707 was characterized, showing that OsMBD707 is constitutively expressed and localized in the nucleus. Transgenic rice overexpressing OsMBD707 exhibited larger tiller angles and altered photoperiod sensitivity, leading to delayed flowering under short day conditions and early flowering under long day conditions. RNA-seq analysis revealed that overexpression of OsMBD707 resulted in reduced photoperiod sensitivity in rice and changes in the expression of flowering regulator genes in the Ehd1-Hd3a/RFT1 pathway.

MBD5 and MBD6 couple DNA methylation to gene silencing through the J-domain protein SILENZIO

Lucia Ichino et al.

Summary: The study identified two Arabidopsis thaliana methyl-CpG-binding domain proteins, MBD5 and MBD6, which are recruited to chromatin by recognition of CG methylation and redundantly repress a subset of genes and transposons without affecting DNA methylation levels. These methyl readers recruit a J-domain protein, SILENZIO, which functions as a transcriptional repressor downstream of DNA methylation. The conserved J-domain motif of SILENZIO is required for its interaction with HSP70s and its silencing function.
