The diversity and breadth of cancer cell fatty acid metabolism

Shilpa R. Nagarajan et al.

Summary: Tumor cellular metabolism has distinguishing features that enhance biomass synthesis and maintain cellular homeostasis through complex interactions between cell-intrinsic and cell-extrinsic factors. Fatty acid metabolism plays a crucial role in tumorigenesis by supporting processes like membrane biosynthesis, energy production, and signaling intermediates generation.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Overcoming enzalutamide resistance in metastatic prostate cancer by targeting sphingosine kinase

Hui-Ming Lin et al.

Summary: This study focused on investigating the association of ceramide-sphingosine-1-phosphate signalling axis with ARSI resistance in mCRPC. Elevated circulating ceramide levels were found to be associated with shorter radiological progression-free survival and overall survival in men with mCRPC. Combining AR aberrations with elevated ceramide levels resulted in a worse prognosis, and SPHK inhibitors showed potential to enhance the efficacy of enzalutamide.

Relationship between Circulating Lipids and Cytokines in Metastatic Castration-Resistant Prostate Cancer

Hui-Ming Lin et al.

Summary: The study found that in metastatic castration-resistant prostate cancer (mCRPC), blood lipids may be more informative than cytokines, with certain lipids consistently associated with poorer clinical outcomes and influencing immune response and treatment response. The correlation between circulating ceramides and cytokines suggests the regulation of immune responses by ceramides, and further research into metabolic interventions related to changes in lipid profiles is warranted.

Aberrations in circulating ceramide levels are associated with poor clinical outcomes across localised and metastatic prostate cancer

Hui-Ming Lin et al.

Summary: Elevated circulating ceramide species are associated with poorer clinical outcomes in prostate cancer patients, and these circulating lipid profiles could be used to predict therapeutic efficacy at different stages of PC.

RB/E2F1 as a Master Regulator of Cancer Cell Metabolism in Advanced Disease

Amy C. Mandigo et al.

Summary: This study identifies the stage-specific consequences of RB loss across cancer progression, which directly impact tumor response to clinically utilized therapeutics. It is the first to investigate the effect of RB loss on global metabolic regulation and link RB/E2F1 to redox control in multiple advanced diseases.

Plasma Cell-Free DNA Profiling of PTEN-PI3K-AKT Pathway Aberrations in Metastatic Castration-Resistant Prostate Cancer

Edmond M. Kwan et al.

Summary: CNVs in the PTEN-PI3K-AKT pathway were readily detected using a novel cfDNA assay in mCRPC patients, with high prevalence of PTEN loss and PIK3CA gain. Additional PTEN-PI3K-AKT pathway aberrations were found in one fifth of PTEN-neutral cases, and concurrent CNVs in the PTEN-PI3K-AKT and AR pathways predicted poor survival outcomes, suggesting potential benefits of dual AR/Akt inhibition in precision treatment for mCRPC.
