Evolutionary and reverse engineering to increase Saccharomyces cerevisiae tolerance to acetic acid, acidic pH, and high temperature

Prisciluis Caheri Salas-Navarrete et al.

Summary: This study used adaptive laboratory evolution (ALE) and whole-genome analysis to generate strains of Saccharomyces cerevisiae tolerant to high temperatures, acetic acid, and acidic pH. Through a year of evolution, two strains were selected, TAT12 and AT22, which were capable of growing and producing ethanol in conditions previously inhibitory to their ancestors. Whole-genome sequencing and transcriptomic analysis showed mutations in key genes related to stress response and replication, confirming the importance of these mutations in enhancing the tolerance of S. cerevisiae to harsh environments.

Summary: This study aimed at developing industrial S. cerevisiae strains with enhanced tolerance towards pretreatment-derived microbial inhibitors by identifying novel gene combinations. The integration of selected genes was proven to increase tolerance to inhibitors in synthetic or industrial hydrolysates, but had a limited impact on xylose fermentation. Certain gene combinations had a reduced impact on xylose conversion.

Summary: By engineering Saccharomyces cerevisiae, near-parity production can be achieved between inhibitor-laden and inhibitor-free feedstocks, and a single strain can be enhanced to tolerate diverse highly toxified feedstocks. Moreover, a lightweight design allows for seamless transferability to existing metabolically engineered chassis strains, demonstrating the potential for cost-effective, at-scale biomass utilization for cellulosic fuel and nonfuel products.
