Summary: Identifying nonnative trimeric forms of SOD1 as toxic species, rather than large aggregates, has changed our understanding of ALS pathophysiology. Large protein aggregates, previously thought to be the central cause of neurodegeneration, are now seen as having a protective role. Understanding the formation and role of nonnative trimers in cell death is key to developing new therapeutics for ALS and other neurodegenerative diseases.
