Brain aging in major depressive disorder: results from the ENIGMA major depressive disorder working group

Laura K. M. Han et al.

Summary: Major depressive disorder is associated with increased risk of brain atrophy, aging-related diseases, and mortality. This study found that MDD patients showed a higher brain-predicted age difference compared to controls, but this difference did not seem to be driven by specific clinical characteristics. Subtle patterns of age-related structural brain abnormalities in MDD were observed, highlighting the need for further longitudinal studies to assess the clinical value of these findings.

The Psychopathology and Neuroanatomical Markers of Depression in Early Psychosis

Rachel Upthegrove et al.

Summary: The study found similarities in symptoms and neuroanatomical features between recent-onset psychosis patients and depression patients, but there was no distinct subgroup of depressed individuals in first-episode psychosis. Depression may be an intrinsic feature of early-stage psychotic disorders, suggesting that treating depression could have widespread benefits.

Summary: The study suggests potential causal associations between IL-6 and schizophrenia as well as depression, and highlights the need for replication in larger samples. Some findings did not survive correction for multiple testing, emphasizing the importance of further research in this area.

Summary: Recent research indicates that schizophrenia may be associated with a noninflammatory synaptic remodeling phenotype of microglia, disrupting neurodevelopment and potentially leading to recurrent episodes of the disorder.

Summary: This study aimed to identify prototypes of depression and psychosis by analyzing clinical data and brain structure data, finding that patients with affective comorbidity were more likely positioned closer to the depression prototype rather than the psychotic prototype.

Brain morphology is differentially impacted by peripheral cytokines in schizophrenia-spectrum disorder

Liliana Laskaris et al.

Summary: The study found differential relationships between peripheral inflammatory markers and select brain regions in first-episode psychosis (FEP) and healthy controls, although no relationships were detected in chronic schizophrenia. Specifically, frontal thickness was positively associated with certain cytokine levels in the healthy control group, while pro-inflammatory cytokines were associated with lower total cortical volume in the FEP group. Longitudinal investigations are needed to determine how the relationship between brain structure and peripheral inflammation may change over time.

Interrelationships between depressive symptoms and positive and negative symptoms of recent onset schizophrenia spectrum disorders: A network analytical approach

Sarah E. Herniman et al.

Summary: This study aimed to understand the interrelationships between positive and negative symptoms of recent-onset schizophrenia spectrum disorders and co-occurring depressive symptoms. Results showed that positive symptoms were associated with depressive and negative symptoms, while negative symptoms were more likely to be associated with depressive symptoms. Suspiciousness and stereotyped thinking bridged between positive and depressive symptoms, and apparent sadness and lassitude bridged between negative and depressive symptoms.

The Impact of Anticholinergic Burden on Functional Capacity in Persons With Schizophrenia Across the Adult Life Span

Waqas Ullah Khan et al.

Summary: The study found that individuals with schizophrenia are commonly exposed to severe anticholinergic burden (ACB), which can have a direct negative impact on functional capacity after controlling for its impact on cognition. ACB, age, education, and cognition were independent predictors of functional capacity, with ACB predicting cognition in those aged 55 years and older. Cognition partially mediated the effect of ACB on functional capacity in this older cohort.
