Summary: Newly designed and synthesized compounds with benzimidazole/benzothiazole structures showed potential antitumor activity by inhibiting proliferation of human lung cancer cell lines. Benzothiazole derivatives exhibited higher activity compared to benzimidazole derivatives. Some compounds also demonstrated antimicrobial activity.

However, certain candidate derivatives exhibited distinct cytotoxicity levels and varying ER alpha affinity correlations, suggesting that the sulfonyl side chain modifications might influence ER alpha binding levels. Overall, the results demonstrated the potential anticancer effects and estrogen receptor modulatory actions of the new methylsulfonyl compounds.

Summary: A new series of benzoxazoles were synthesized as potential anticancer agents targeting SIRT1 for NSCLC. Compounds 3e and 3g showed strong anticancer effects on A549 cells, inducing apoptosis in a dose-dependent manner. In vitro and in silico studies indicated that these compounds could be promising orally bioavailable agents for targeted therapy of NSCLC.

Summary: Benzimidazol-2-yl)-7-hydroxycoumarin and N-(2-hydroxyphenyl)-7-hydroxycoumarin-3-carboxamide were synthesized through condensation in acetic acid, followed by acetylation and halogenation. The synthesized molecules showed good in vitro antiproliferative activities against breast cancer, particularly 3-(benzimidazol-2-yl)-7-acetoxycoumarin which exhibited a significant cytotoxic effect compared to the standard drug DOX (doxorubicin).
