Impact of the Expression System on Recombinant Protein Production in Escherichia coli BL21

Summary: The research evaluated the expression of YFP reporter protein under the control of different strength promoters and replication origins, with different E. coli strains and carbon sources. It was found that metabolic burden leads to decreased expression of recombinant proteins, highlighting the importance of balancing plasmid copy number and promoter strength for optimal expression. The results represent a significant advance in determining the most suitable expression system for improving both the quantity and quality of recombinant proteins in bioproduction engineering.

Metabolic and physiological changes in host cells caused by heat stress and overproduction of recombinant proteins are described, along with discussions on the current status and future applications of systems using heat to induce protein production.
