Seconds-Resolved, In Situ Measurements of Plasma Phenylalanine Disposition Kinetics in Living Rats

Andrea Idili et al.

Summary: This study utilized real-time EAB sensors to monitor plasma phenylalanine levels in live animals, revealing previously unreported metabolic response phases. By analyzing hundreds of individual measurements, inter-subject variability, including significant differences related to feeding status, was identified. This approach has significant implications for understanding physiology and monitoring and treating metabolic disorders.

Interrogation of Electrochemical Aptamer-Based Sensors via Peak-to-Peak Separation in Cyclic Voltammetry Improves the Temporal Stability and Batch-to-Batch Variability in Biological Fluids

Miguel Aller Pellitero et al.

Summary: This study demonstrates the potential advantages of using cyclic voltammetry to directly measure electron-transfer changes, reporting target concentration through changes in the peak-to-peak separation, ΔE-P, of cyclic voltammograms. Sensors interrogated with ΔE-P are resistant to drift, showing decreased variability in performance between batches and days. Additionally, ΔE-P-based measurements can be performed quickly and are competitive with other subsecond interrogation strategies.

Chemical Equilibrium-Based Mechanism for the Electrochemical Reduction of DNA-Bound Methylene Blue Explains Double Redox Waves in Voltammetry

J. D. Mahlum et al.

Summary: Methylene blue is commonly used as a redox reporter in DNA-based electrochemical sensors, with recent research focusing on its use in aptamer-based sensors. A numerical model was developed to analyze the redox waves observed at higher scanning rates, revealing the protonation equilibrium of the radical intermediate species formed during electron transfer. This model also showed the impact of blocking monolayer chemistries on electron-transfer kinetics in these sensors.

Elucidating the Mechanisms Underlying the Signal Drift of Electrochemical Aptamer-Based Sensors in Whole Blood

Kaylyn K. Leung et al.

Summary: The development of electrochemical aptamer-based sensors has enabled real-time monitoring of specific molecules in vivo, but there is still a need to address signal drift for longer in vivo deployments. Research has identified electrochemically driven desorption of self-assembled monolayers and fouling by blood components as the main sources of signal loss in challenging in vivo conditions.
