The long noncoding RNA glycoLINC assembles a lower glycolytic metabolon to promote glycolysis

Youming Zhu et al.

Summary: This study reveals a novel long noncoding RNA (lncRNA) called gLINC that acts as a backbone for the formation of a metabolon involving glycolytic enzymes and lactate dehydrogenase A. The gLINC metabolon enhances glycolytic flux, ATP production, and cell survival under serine deprivation. Overexpression of gLINC in cancer cells promotes xenograft growth in mice on a serine-deprived diet, suggesting its role in cancer cell adaptation.

Phosphofructokinase relocalizes into subcellular compartments with liquid-like properties in vivo

SoRi Jang et al.

Summary: This study systematically examined the dynamic subcellular localization of glycolytic protein phospho-fructokinase-1/PFK-1.1 in Caenorhabditis elegans, showing that PFK-1.1 can form condensates near synapses in neurons in response to energy stress and exhibit liquid-like properties. The findings suggest that PFK-1.1 condensates may represent novel metabolic subcompartments.

Relocation of phosphofructokinases within epithelial cells is a novel event preceding breast cancer recurrence that accurately predicts patient outcomes

Richard A. Cheung et al.

Summary: This study analyzed the differences in PFKL and PFKFB4 distribution within ductal epithelial cells from breast cancer samples, showing potential for predicting cancer recurrence risk. Machine learning was utilized to accurately predict patient outcomes based on these biomarkers. The PFK enzyme patterns within epithelial cells were identified as metabolic switches representing key mechanistic steps of cancer recurrences.

Increased demand for NAD+ relative to ATP drives aerobic glycolysis

Alba Luengo et al.

Summary: Increasing the activity of the pyruvate dehydrogenase complex impairs cell proliferation by reducing the NAD(+)/NADH ratio, which is caused by increased mitochondrial membrane potential. When demand for NAD(+) exceeds the demand for ATP in cells, aerobic glycolysis is promoted despite available oxygen.

ATM inhibitor KU-55933 induces apoptosis and inhibits motility by blocking GLUT1-mediated glucose uptake in aggressive cancer cells with sustained activation of Akt

Benjamin R. E. Harris et al.

Summary: Enhanced glucose uptake in cancer cells is closely correlated with poor prognosis, and the protein kinase ATM plays a key role in insulin-mediated glucose uptake. Inhibition of ATM by KU-55933 inhibits glucose uptake and glycolysis, induces apoptosis, and inhibits motility of cancer cells with overactivated Akt, showing potential as a therapeutic agent against cancer.
