Identification of a novel PHGDH covalent inhibitor by chemical proteomics and phenotypic profiling

Summary: This study identifies withangulatin A (WA) as a novel covalent inhibitor of phosphoglycerate dehydrogenase (PHGDH), the first rate-limiting enzyme of the serine synthesis pathway. WA directly binds to PHGDH and inactivates its enzyme activity by covalent modification of cysteine 295 residue (Cys295). Inhibition of PHGDH by WA leads to decreased glutathione synthesis and increased levels of reactive oxygen species (ROS), resulting in inhibition of tumor proliferation.
