Genetic and functional analysis of KIF5A variants in Japanese patients with sporadic amyotrophic lateral sclerosis

Summary: Two recent genetic studies found that loss-of-function mutation of the C-terminal cargo-binding tail domain of the KIF5A gene cause amyotrophic lateral sclerosis (ALS). This study in Japanese patients with sporadic ALS identified a novel loss-of-function variant in KIF5A, highlighting its rare occurrence as a cause of ALS.

Microtubule Stabilization Promotes Microcirculation Reconstruction After Spinal Cord Injury

Summary: The study demonstrates that microtubule stabilization treatment can improve microcirculation reconstruction after spinal cord injury by increasing the number of microvessels, pericytes, and perfused microvessels, while also reducing apoptosis and migration of endothelial cells and pericytes and promoting their proliferation and survival. This suggests the potential of microtubule stabilization as a therapeutic target to reduce microcirculation dysfunction after SCI in the clinic.

Optic nerve regeneration screen identifies multiple genes restricting adult neural repair

Summary: Adult mammalian central nervous system (CNS) trauma leads to persistent neurological deficits due to minimal axonal regeneration. In this study, suppression of 40 genes was found to promote retinal ganglion cell axon regeneration after optic nerve crush. Loss of interleukin 22 (IL-22) contributes to early inflammatory responses, activation of Stat3, DLK pathways, and upregulation of neuron-intrinsic regeneration-associated genes.

Ultrasonic control of neurite outgrowth direction

Summary: This study found that ultrasound vibration can control the direction and length of neurite outgrowth, with neurites orienting circumferentially under ultrasonication and the spatial gradient of ultrasound vibration affecting neurite growth direction. Ultrasonication with 30 V-pp for 72 h increased neurite length by 99.7% compared to unsonicated cells.

Additionally, α-TubK40me3 potently promotes tubulin nucleation and is involved in tubulin acetylation regulation, highlighting its importance in microtubule dynamics and neuronal development.

Topoisomerase I inhibition and peripheral nerve injury induce DNA breaks and ATF3-associated axon regeneration in sensory neurons

Summary: This study shows that topoisomerase inhibitors, including camptothecin, can induce ATF3 expression, promote neurite outgrowth in sensory neurons, and enhance axonal regeneration. DNA breaks occur immediately after nerve damage at the ATF3 gene locus in injured sensory neurons and are further increased after camptothecin exposure, potentially contributing to the initiation of transcriptional changes required for peripheral nerve regeneration.
