Discovery of cellular substrates of human RNA-decapping enzyme DCP2 using a stapled bicyclic peptide inhibitor

Summary: DCP2 is an RNA-decapping enzyme that regulates the stability of human RNAs encoding factors involved in transcription and the immune response. In this study, a cell-permeable and selective chemical inhibitor of DCP2, CP21, was identified and shown to inhibit DCP2 decapping activity and increase the formation of P-bodies in human cells. The inhibition of DCP2 by CP21 led to the discovery of 76 previously unreported DCP2 substrates, demonstrating the potential of DCP2 inhibition as a complement to genetic approaches in studying RNA decay.
