Insulin-dependent glucose consumption dynamics in 3D primary human liver cultures measured by a sensitive and specific glucose sensor with nanoliter input volume

Summary: A colorimetric glucose assay based on the glucose oxidase-peroxidase system was developed for monitoring glucose consumption dynamics in 3D primary human liver cell cultures. The system showed high sensitivity and accuracy, and could be integrated into microfluidic devices for longitudinal profiling of hepatic glucose consumption. Studies using this system found that insulin and extracellular glucose levels accounted for a significant portion of hepatic glucose consumption.

Primary Human Hepatocyte Spheroids as an In Vitro Tool for Investigating Drug Compounds with Low Hepatic Clearance

Summary: This study demonstrated the accurate prediction of hepatic clearance for slowly metabolized drug compounds using primary human hepatocyte spheroids. The spheroids maintained their in vivo-like phenotype during prolonged incubations, allowing for monitoring of parent drug depletion for up to 7 days. However, attempts to increase metabolic capacity by pooling hepatocyte spheroids resulted in fusion and de-differentiation processes, showing limited applicability for quantitative pharmacokinetic studies.
