Stereoselective Organocatalytic Construction of Spiro Oxindole Pyrrolidines Using Unsaturated α-Ketoesters and α-Ketoamides

Summary: The stereoselective formation of spiro oxindole pyrrolidines via formal [3+2] cycloaddition of oxindole imines with ketoesters and ketoamides was investigated, with bifunctional squaramide organocatalyst inducing enantioselectivity of up to 72% ee. Ketoamides provided alternative products, and the reaction pathway was elucidated with the help of DFT calculation.

Construction of Chiral Isotetronic Acid-Fused Thiochromane via Doubly Annulative Strategy

Summary: A sulfa-Michael/aldol/lactonization cascade reaction has been established to construct isotetronic acid-fused thiochromanes in a highly stereoselective fashion (>= 11:1 dr, 35-98% ee). The tricyclic products were obtained in 35-99% isolated yields in the presence of a bifunctional squaramide. Three reactive sites of beta,gamma-unsaturated alpha-ketoester, including the less-explored ester carbonyl group, were sequentially utilized to construct two fused heterocycles in a one-pot operation.

Organocatalytic Asymmetric Synthesis of Aza-Spirooxindoles via Michael/Friedel-Crafts Cascade Reaction of 1,3-Nitroenynes and 3-Pyrrolyloxindoles

Summary: By using a chiral bifunctional squaramide catalyst, an asymmetric [3+3] cyclization of nitroenynes and 3-pyrrolyloxindoles has been achieved, providing an efficient access to enantioenriched polycyclic aza-spirooxindoles with 32-95% isolated yields and excellent stereocontrol under mild reaction conditions.
