Summary: The NPY family is a peptide-activated G protein-coupled receptor system involved in various biological processes. Comparing the NPY system in evolutionarily ancient organisms reveals conserved structural determinants of peptide recognition and receptor activation. Common features include acidic patches in the extracellular loops of the receptor, along with a structurally conserved glutamate that anchors peptides via a salt bridge to an arginine residue. Variability in peptide binding is enabled by receptor-specific interactions, with a family-conserved residue Q3.32 playing a key role in peptide binding and receptor activation.
