Efficient L-Proline catalyzed synthesis of some new (4-substituted-phenyl)-1,5-dihydro-2H-pyrimido[4,5-d][1,3]thiazolo[3,2a]-pyrimidine-2,4(3H)-diones bearing thiazolopyrimidine derivatives and evaluation of their pharmacological activities

S. H. Sukanya et al.

Summary: In this study, a simple and efficient method was developed to synthesize a series of new compounds, which were evaluated for in vitro cytotoxicity and anti-inflammatory effects. The results indicated that some compounds showed potential cytotoxic effects and significant anti-inflammatory effects, and the structure-activity relationship study highlighted the influence of electron-withdrawing groups on compound activity.

Inhibition of ubiquitin-specific protease 7 sensitizes acute myeloid leukemia to chemotherapy

Maelle Cartel et al.

Summary: The research demonstrates that inhibition of deubiquitinylase USP7 reduces cell proliferation, blocks DNA replication, and increases cell death in AML. High expression of USP7 is associated with resistance to therapy in AML patients, indicating USP7 as both a marker of resistance and a potential therapeutic target for overcoming treatment resistance.

A Dual-Acting Nitric Oxide Donor and Phosphodiesterase 5 Inhibitor Promotes Wound Healing in Normal Mice and Mice with Diabetes

Maya Ben-Yehuda Greenwald et al.

Summary: TOP-N53, a compound with dual-acting activity as a nitric oxide donor and phosphodiesterase 5 inhibitor, demonstrates remarkable healing-promoting effects by promoting keratinocyte proliferation, angiogenesis, and collagen maturation in healthy mice and partially rescuing healing impairment in mice with genetically determined type II diabetes.

Mechanisms of stearoyl CoA desaturase inhibitor sensitivity and acquired resistance in cancer

Nicole Oatman et al.

Summary: In glioblastoma, some patients have low levels of SCD expression, leading to the evolution of cell lines that can survive independent of SCD, despite unknown mechanisms. Cell lines that escape genetic and epigenetic alterations become highly dependent on SCD for survival. Resistance to SCD inhibition in SCD-dependent lines is mediated through a previously unknown FOSB mechanism, which can be blunted to extend survival in tumor-bearing mice.
