Cleavage of DNA and RNA by PLD3 and PLD4 limits autoinflammatory triggering by multiple sensors

Quantification and characterization of the 5 ' exonuclease activity of the lysosomal nuclease PLD3 by a novel cell-based assay

Summary: Phospholipase D3 (PLD3) and phospholipase D4 (PLD4) are recently described lysosomal nucleases associated with various diseases. A cell-based assay was developed to quantify the 5' exonuclease activity of PLD3 and PLD4, revealing PLD3 as the primary acid 5' exonuclease with higher specific activity in HeLa cells. Experiments on genetic variants of PLD3 found that some variants result in reduced activity.
