A novel SPE-UHPLC-DAD method for the determination of fumagillin produced by Aspergillus fumigatus in cell culture media

Summary: Fumagillin, produced by Aspergillus fumigatus, is gaining importance due to its relevance to invasive aspergillosis. A validated method for quantitative analysis of fumagillin in RPMI-1640 has been developed, meeting validation criteria for bioanalysis and offering a suitable limit of quantitation. The method was successfully applied to quantifying fumagillin produced by different strains of Aspergillus fumigatus.

Determining Aspergillus fumigatus transcription factor expression and function during invasion of the mammalian lung

Summary: Although the majority of genes in the A. fumigatus genome have unknown functions, this study identified two transcription factors, RlmA and SltA, as crucial for virulence. The organism responds to nutrient limitation and stress by up-regulating genes encoding mycotoxins and secondary metabolites during growth in the lungs. Invasive growth in vivo induces a unique transcriptional profile as the organism faces nutrient limitation and attacks by host phagocytes, similar to growth in in vitro medium deficient in nitrogen, iron, and/or zinc.
