Enhancement of osteoporotic fracture healing by vibration treatment: The role of osteocytes

Wing Hoi Cheung et al.

Summary: The prevalence of osteoporotic fracture is high due to global aging, and delayed healing increases socioeconomic burden significantly. Mechanical stimulation, particularly vibration treatment, has shown promising results in enhancing osteoporotic fracture healing by improving callus formation, mineralization, and remodeling. Osteocytes play a key role in sensing mechanical signals and facilitating healing, with vibration treatment significantly increasing mineralization and accelerating fracture healing. Further studies and randomized controlled trials are needed to validate the efficacy of vibration treatment for osteoporotic fracture patients.

Combination of magnesium ions and vitamin C alleviates synovitis and osteophyte formation in osteoarthritis of mice

Hao Yao et al.

Summary: The combination therapy of magnesium ions and vitamin C significantly alleviated joint destruction and pain in osteoarthritis, with sustained effects for up to 3 months. Vitamin C enhanced the promotive effect of magnesium ions on HIF-1 alpha expression in cartilage, and the combination also promoted M2 polarization of macrophages in the synovium.

High-purity magnesium pin enhances bone consolidation in distraction osteogenesis via regulating Ptch protein activating Hedgehog-alternative Wnt signaling

Musha Hamushan et al.

Summary: Magnesium alloys have shown potential as biomaterials for orthopedic implants due to their degradability, osteogenic effects, and biocompatibility. By developing and using high-purity magnesium pins in distraction osteogenesis, it was found that they can promote bone healing and shorten the consolidation time. Further investigation revealed that magnesium may regulate Ptch protein to activate the Hedgehog-alternative Wnt signaling pathway.

Synergistic effects of magnesium ions and simvastatin on attenuation of high-fat diet-induced bone loss

Bingyang Dai et al.

Summary: The combination of magnesium and simvastatin showed synergistic effects in attenuating high-fat diet-induced bone loss in mice by inhibiting osteoclast formation and bone resorption, reducing hepatic cholesterol synthesis, and inhibiting matrix metallopeptidase 13 (Mmp13) expression in pre-osteoclasts. This combination therapy may be a cost-effective alternative treatment for obesity-related bone loss.

Emerging insights into the comparative effectiveness of anabolic therapies for osteoporosis

Eben G. Estell et al.

Summary: The mainstay treatment for osteoporosis has traditionally been antiresorptive agents, but declining adherence has led to a plateau in fracture rates reduction. The approval of anabolic agents in the past two decades represents a new approach to improving bone quality and reducing fracture risk, potentially leading to a new clinical paradigm.
