Spatiotemporal manipulation of L-arginine release from bioactive hydrogels initiates rapid skin wound healing accompanied with repressed scar formation

Summary: In this study, biofunctional hydrogel (CA) was synthesized by introducing L-arginine (L-Arg) into chitosan (CS) for spatial and temporal manipulation of L-Arg release kinetics, leading to enhanced wound healing and reduced scar formation. Experiments and simulations showed that different substitution degrees of L-Arg on CS influenced their molecular arrangements and interactions, affecting L-Arg release profiles. An optimized hydrogel with 8.50% substitution degree (CA2) was found to promote wound healing and reduce scar formation in both in vitro and in vivo studies.
