Salinomycin mediated therapeutic targeting of circulating stem like cell population in oral cancer

Shanaya Patel et al.

Summary: Salinomycin was identified as a potential lead compound that effectively inhibits CD44 receptor in circulating tumor stem cells (CTSCs), demonstrating significant cytotoxic effect towards the CD44+ subpopulation in a dose and time dependent manner. This study showed the potential of Salinomycin to target chemo-resistant circulating CD44 population by attenuating its proliferation and survival.

Inhibition of the Lipid Droplet-Peroxisome Proliferator-Activated Receptor α Axis Suppresses Cancer Stem Cell Properties

Kenta Kuramoto et al.

Summary: This study found that the accumulation of LDs in certain cancer stem cells leads to the activation of PPAR alpha, and the metabolic turnover of LDs accompanying PPAR alpha activation helps maintain the properties of cancer stem cells. Inhibition of PPAR alpha, re-esterification, and lipolysis pathways can suppress cancer stemness.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Diagnosis and Treatment of Metastatic Colorectal Cancer: A Review

Leah H. Biller et al.

Summary: Colorectal cancer is the third most common cause of cancer mortality worldwide. For patients diagnosed with metastatic colorectal cancer, approximately 70% to 75% can survive beyond 1 year. Tailoring treatments based on the molecular and pathologic features of the tumor has shown to improve overall survival rates.

Summary: PEITC may have the potential to prevent or delay colorectal cancer growth by inhibiting the properties of CSCs, especially clonogenicity and the expression of pluripotent factors. RNA-sequencing results demonstrated that PEITC pre-treatment significantly suppressed genes related to inflammatory, immune responses, and chemokine-related signaling.

PIK3C3 Inhibition Promotes Sensitivity to Colon Cancer Therapy by Inhibiting Cancer Stem Cells

Balawant Kumar et al.

Summary: Colorectal cancer (CRC) therapy faces challenges due to cancer stem cells (CSCs) resistance to chemotherapy. Inhibiting autophagy with 36-077, a potent PIK3C3/VPS34 inhibitor, shows promise in improving the efficacy of CRC therapy by targeting CSCs.

USP47 maintains the stemness of colorectal cancer cells and is inhibited by parthenolide

Shaohua Zhang et al.

Summary: The upregulation of USP47 is associated with tumorigenesis and poor prognosis in clinical patients with colorectal cancer and is closely involved in the maintenance of the stemness of colorectal cancer stem cells (CCSCs). Silencing USP47 reduces proliferation and migration of colorectal cancer cells, suppresses the self-renewal of CCSCs, and induces apoptosis of CCSCs. Parthenolide (PTL) as a novel USP47 inhibitor shows potential in targeting CCSCs for colorectal cancer treatment.

Honokiol Affects Stem Cell Viability by Suppressing Oncogenic YAP1 Function to Inhibit Colon Tumorigenesis

Dharmalingam Subramaniam et al.

Summary: Honokiol (HNK) inhibits proliferation and colony formation, induces apoptosis, and suppresses colonosphere formation in colon cancer cells. HNK interacts with DCLK1, suppresses its kinase activity, and inhibits the expression of cancer stem cell marker proteins such as LGR5 and CD44. Additionally, HNK prevents colon tumorigenesis by inducing PUMA-YAP1 interaction and cytoplasmic sequestration, thereby suppressing oncogenic YAP1 activity.
