Pathological mechanisms and therapeutic strategies for Alzheimer's disease

Yaojun Ju et al.

Summary: Alzheimer's disease is a complex neurodegenerative disease characterized by synaptic dysfunctions and neurotransmitter deficiencies. The pathological features of Alzheimer's disease include changes in β-amyloid and tau proteins. Aging and other factors are also risk factors for the disease. Therefore, developing treatment strategies that target different pathological factors is crucial.

The diversity and composition of the human gut lactic acid bacteria and bifidobacterial microbiota vary depending on age

Teng Ma et al.

Summary: The study found that as individuals age, changes occur in the diversity and composition of gut bifidobacterial microbiota, with a negative correlation observed with age and alpha diversity in these subpopulations. The structure and composition of gut bifidobacterial subpopulation in longevous elderly group were different from other age groups, while the gut LAB subpopulation in young adults significantly differed from all three elderly groups. Several potentially beneficial bacteria were identified in the longevous elderly group, with the relative abundance of Bifidobacterium adolescentis decreasing significantly with age. The age-dependent distribution of bifidobacteria and LAB varied, suggesting different life stage changes and functional roles.

Lactobacillus rhamnosus GG Colonization in Early Life Ameliorates Inflammaging of Offspring by Activating SIRT1/AMPK/PGC-1α Pathway

Tianyu Liu et al.

Summary: Research has shown that early-life colonization of Lactobacillus rhamnosus GG (LGG) can ameliorate inflammaging in offspring by altering gut microbiota composition, increasing abundance of longevity-related bacteria, promoting short-chain fatty acid production, enhancing antioxidant defense, and inhibiting intestinal low-grade inflammation. Mechanistically, LGG upregulates the SIRT1/AMPK/PGC-1 alpha pathway and suppresses NF-kappa B activation to protect against inflammaging, providing a new strategy for preventing age-related diseases.

The Role of Gut Bacterial Metabolites in Brain Development, Aging and Disease

Shirley Mei-Sin Tran et al.

Summary: Emerging evidence suggests correlations between gut microbiome and human health, including brain-related diseases. This study systematically assessed literature on gut bacterial metabolites and their associations with diseases of the central nervous system. Findings indicate altered bacterial metabolites in various brain diseases, with potential neuroactive effects by certain gut-derived metabolites.

The pleiotropic effects of prebiotic galacto-oligosaccharides on the aging gut

Jason W. Arnold et al.

Summary: Dietary GOS had a positive impact on the microbiome composition and intestinal health of both old and young animals, but a consistent bifidogenic effect was not observed. Antibiotics had a more significant impact on regulating inflammation markers, while GOS improved the intestinal barrier homeostasis and increased mucus production to promote gut health in the elderly.

AMPK: a key regulator of energy stress and calcium-induced autophagy

Rimpi Saikia et al.

Summary: Autophagy is a cell survival strategy that helps cells maintain homeostasis by degrading and reusing cellular components under unfavorable conditions. It is regulated by AMPK and mTORC1, with AMPK activating autophagy in energy-deprived conditions and mTORC1 inhibiting autophagy in nutrient-rich conditions. Additionally, cytoplasmic calcium can also activate AMPK and autophagy through a calmodulin/CaMKK beta pathway, although the physiological significance of this regulation is still unclear.

Gut microbial metabolite urolithin B attenuates intestinal immunity function in vivo in aging mice and in vitro in HT29 cells by regulating oxidative stress and inflammatory signalling

Peng Chen et al.

Summary: Urolithin B (Uro B) has been shown to possess antioxidant and anti-inflammatory effects, with potential to ameliorate intestinal immunity function and protect against aging-related injuries in mice. This compound could be considered as a healthcare product for preventing diseases associated with immune system aging.
