N-Alkylpyridinium sulfonates for retention time indexing in reversed-phase-liquid chromatography-mass spectrometry-based metabolomics

Rainer Stoffel et al.

Summary: Chromatographic retention time information is crucial for metabolite identification, but variations in retention times can occur due to factors like column dead volumes and gradient dwell volume. A new universal retention time indexing system based on N-alkylpyridinium sulfonates has been developed for liquid chromatography, showing potential for improving comparability and enabling metabolite annotation and identification. This system has the advantage of being applicable to both positive and negative ionization modes, with UV activity, making it useful for reversed-phase liquid chromatography-mass spectrometry-based metabolomics.

High-confidence structural annotation of metabolites absent from spectral libraries

Martin A. Hoffmann et al.

Summary: The COSMIC workflow combines in silico structure database generation and annotation with a confidence score for accurate and efficient metabolite annotation. It outperforms spectral library search by annotating a substantial number of hits at low false discovery rates and can successfully annotate structures never reported before.

metID: an R package for automatable compound annotation for LC-MS-based data

Xiaotao Shen et al.

Summary: Accurate and efficient compound annotation has been a challenge for LC-MS-based data, but efforts to overcome this obstacle have been limited by the sources of spectral information used. The metID R package aims to address this issue by combining information from major databases for streamlined compound annotation, making the process automatic and reproducible across platforms.

Development and Application of Liquid Chromatographic Retention Time Indices in HRMS-Based Suspect and Nontarget Screening

Reza Aalizadeh et al.

Summary: This study proposed a unified retention time index (RTI) approach for liquid chromatography, aiming to achieve comparable retention times among different laboratories. By calibrating the elution pattern with two sets of calibration compounds, the RTI range was set between 1 and 1000, which was considered the most appropriate RTI system after rigorous evaluation.
