Diagnostic biomarkers from proteomic characterization of cerebrospinal fluid in patients with brain malignancies

Summary: Recent advancements in molecular diagnostics using liquid biopsies have shown promise in monitoring tumor evolution and treatment response for brain malignancies. A study utilizing mass spectrometry-based protein analysis of cerebrospinal fluid samples identified potential biomarkers for glioblastoma and revealed proteomic changes associated with different brain tumor entities. This approach has the potential to enhance diagnosis and therapy monitoring for brain malignancies.

Summary: The study found that levels of CNS-specific proteins brevican and neurocan did not differ between lumbar and ventricular CSF. However, several CNS-unspecific MMPs and TIMP-1 were significantly increased in lumbar CSF, suggesting potential contribution from peripheral tissues. The increase of ECM proteins in CSF following shunt surgery may indicate disrupted ECM dynamics in iNPH that are restored by restitution of CSF dynamics.

Tumor DNR Mutations From Intraparenchymal Brain Metastases Are Detectable in CSF

Summary: This study investigated the use of cell-free DNA (cfDNA) in cerebrospinal fluid (CSF) as a surrogate marker for genomic profiling of intraparenchymal (IP) brain metastases. It found cancer-associated mutations in CSF of patients with IP brain metastases, suggesting that CSF analysis can be an alternative method to detect somatic mutations in brain metastases. Detection of mutational DNA in CSF was not restricted to leptomeningeal disease and was found in two thirds of patients with IP brain metastases in the cohort.
