Summary: Stroke is the second leading cause of death and the third leading cause of disability worldwide, with edema playing a major role in stroke-associated morbidity and mortality. Regulating water balance in the brain through targeting AQP4 channels may be a promising therapeutic approach for treating brain edema. The FDA-approved drug trifluoperazine (TFP) has shown efficacy in reducing cerebral edema in post-stroke mice by inhibiting AQP4 expression, without inducing deleterious changes in brain electrolytes or metabolic markers.
