Tumor-Associated Macrophages in Pancreatic Ductal Adenocarcinoma: Origin, Polarization, Function, and Reprogramming

Sen Yang et al.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is a deadly malignancy that can only be cured by surgery in its early stages, but recurrence remains a high possibility. Tumor-associated macrophages (TAMs) play a crucial role in PDAC development, promoting tumor behaviors such as tumorigenesis and metastasis.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The CDCP1 Signaling Hub: A Target for Cancer Detection and Therapeutic Intervention

Tashbib Khan et al.

Summary: CDCP1 as an important hub for oncogenic signaling is upregulated in various malignancies and can be targeted for cancer diagnosis and therapy. Elevated levels of CDCP1 are associated with disease progression and poorer survival, predominantly located on the cell surface and involved in multiple cancer cell signaling pathways.

Summary: The study found that overexpression of S100A family members is closely related to the pathological features and prognosis of PC, and identified several CpG sites that are correlated with the expression of S100A family members and OS. Through multiple bioinformatic analyses, the study revealed the potential oncological functions of the S100A family in PC.

Alpha-Enolase: Emerging Tumor-Associated Antigen, Cancer Biomarker, and Oncotherapeutic Target

Frankis A. Almaguel et al.

Summary: ENO1 is a glycolytic enzyme that also acts as a plasminogen receptor on the cell surface, contributing to cancer cell proliferation and metastasis. Its overexpression in various cancers is associated with poor prognosis and the generation of anti-ENO1 autoantibodies, making it a potential therapeutic target and cancer biomarker.

Summary: Pancreatic cancer has a poor prognosis, and surgery with adjuvant chemotherapy is the only curative option for a small group of patients. Novel therapeutic strategies are urgently needed for advanced or metastatic cases where surgical resection is not possible. Molecular profiling and targeted therapies show promise in overcoming the limitations of conventional treatments. Currently, personalized and multimodal targeted therapies are undergoing clinical trials with promising results targeting various aspects of pancreatic cancer.

Screening of 5-and 6-Substituted Amiloride Libraries Identifies Dual-uPA/NHE1 Active and Single Target-Selective Inhibitors

Benjamin J. Buckley et al.

Summary: The K+-sparing diuretic amiloride has demonstrated off-target anti-cancer effects by inhibiting the serine protease urokinase-type plasminogen activator (uPA) and the sodium-hydrogen exchanger isoform-1 (NHE1). Through screening, new chemotype-matched, non-cytotoxic probes have been identified as selective uPA inhibitors, dual-targeting inhibitors, and high-NHE1 potency inhibitors for dissecting the pharmacological effects of selective uPA and NHE1 inhibition versus dual-uPA/NHE1 inhibition.

Substrate-biased activity-based probes identify proteases that cleave receptor CDCP1

Thomas Kryza et al.

Summary: The study identifies uPA as the master regulator of CDCP1 proteolysis, acting by directly cleaving CDCP1 and activating plasmin. Co-expression of uPA and CDCP1 predicts poor disease outcome in multiple cancers, and uPA-mediated CDCP1 proteolysis promotes metastasis in clinical models. These results suggest CDCP1 cleavage as a potential target for disrupting cancer, and establish sbABP technology as a novel approach for identifying disease-relevant proteases.
