Zeb2/Axin2-Enriched BMSC-Derived Exosomes Promote Post-Stroke Functional Recovery by Enhancing Neurogenesis and Neural Plasticity

Rui Wei et al.

Summary: In this study, exosomes harvested from BMSCs enriched with Zeb2/Axin2 were found to enhance post-stroke neuroplasticity and functional recovery in MCAO rats by promoting proliferation and differentiation of neural stem cells. The mechanisms may be related to the SOX10, Wnt/β-catenin, and endothelin-3/EDNRB pathways.

MiR-17-92 enriched exosomes derived from multipotent mesenchymal stromal cells enhance axon-myelin remodeling and motor electrophysiological recovery after stroke

Hongqi Xin et al.

Summary: Exosomes enriched with the MiR-17-92 cluster derived from multipotent mesenchymal stromal cells (MSCs) can enhance functional recovery after transient stroke, potentially by promoting axonal extension and myelination. This enhancement in axon-myelin remodeling may be mediated through the activation of the PI3K/Akt/mTOR pathway induced by downregulation of PTEN.

Microglia exosomal miRNA-137 attenuates ischemic brain injury through targeting Notch1

Dianquan Zhang et al.

Summary: This study found that M2 microglial exosomes (BV2-Exo) were internalized by neurons, attenuating neuronal apoptosis in response to ischemic injury and reducing infarct volume and behavioral deficits in ischemic mice. Exosomal miRNA-137 played a role in this neuroprotective effect by targeting Notch1.

Targeted delivery of neural progenitor cell-derived extracellular vesicles for anti-inflammation after cerebral ischemia

Tian Tian et al.

Summary: Ischemic stroke is a major cause of death and anti-inflammatory strategies are promising for reducing brain injury during reperfusion. Stem cell-derived extracellular vesicles (EVs) have emerged as therapeutic agents for immune modulation. By attaching targeting ligands onto EVs using a recombinant fusion protein, researchers were able to deliver anti-inflammatory EVs to the ischemic brain, showing promising results in suppressing inflammation.

Lithium modulates miR-1906 levels of mesenchymal stem cell-derived extracellular vesicles contributing to poststroke neuroprotection by toll-like receptor 4 regulation

Matteo Haupt et al.

Summary: Preconditioning of MSCs with lithium enhances the therapeutic potential of EVs, specifically Li-EVs, which show increased resistance against hypoxic injury and improved neurological recovery in poststroke models. Additionally, Li-EVs modify the contents of miRNAs to inhibit TLR4 signaling pathway and reduce poststroke cerebral inflammation, providing acute neuroprotection and enhanced neurological recovery.
