The placenta-brain-axis

Summary: The placenta is a structurally diverse organ in mammals that not only facilitates exchange of gases and nutrients, but also produces hormones and neurotransmitters influencing maternal physiology and fetal development. Placental responses to in utero stressors may have long-term health outcomes, particularly impacting brain development. This intimate relationship between the placenta and brain has led to the term placenta-brain-axis, which is being studied for its effects on later neurobehavioral disorders.

Transcriptome-wide association analysis of brain structures yields insights into pleiotropy with complex neuropsychiatric traits

Summary: The study found that structural variations of the human brain are highly heritable and have been linked to various complex traits. By performing cross-tissue TWAS analysis on 211 structural neuroimaging, the researchers identified 278 associated genes. Furthermore, TWAS gene-based polygenic risk scores showed significant improvement in association analysis compared to conventional variant-based GWAS PRS, explaining up to 6.97% of phenotypic variance.

The RNA landscape of the human placenta in health and disease

Summary: The study analyzed RNA from 302 human placentas and found that the placental transcriptome has low complexity, with a small number of genes accounting for a large portion of reads. Multiple circular RNAs and novel piRNAs were identified, as well as differences in mRNA, lincRNA, and circRNA associated with PE and FGR. Furthermore, the study identified a secreted protein FSTL3 as being differentially expressed in both PE and FGR, with elevated serum levels predicting subsequent PE and FGR.

Examining Sex Differences in the Human Placental Transcriptome During the First Fetal Androgen Peak

Summary: Sex differences in human placenta exist from early pregnancy to term, with 322 transcripts differentially expressed between male and female CV from 11 to 16 weeks. Contrary to expectations, the difference between male and female expression was more pronounced at earlier gestational ages, with females showing upregulation in extracellular matrix components and X-linked genes, while males exhibited upregulation of transcripts related to chromosome 19. Some of the highly differentially expressed autosomal genes were found to regulate immune cell interactions. This study provides insights into sex-linked gene expression in developing human placenta and sets the foundation for understanding the mechanistic origins of sex differences in prenatal development.
