Unnatural bases for recognition of noncoding nucleic acid interfaces

Shiqin Miao et al.

Summary: The concept of using synthetic heterocycles in place of natural bases to interact with DNA and RNA has been studied for almost 60 years, with potential to expand genetic code and access new molecular functions. While much research focuses on code expansion, there is also increasing effort in docking synthetic heterocycles to noncoding nucleic acid interfaces to elucidate major nucleic acid processes. Both coding and noncoding interface research share fundamental principles in molecular recognition.

RNA origami design tools enable cotranscriptional folding of kilobase-sized nanoscaffolds

Cody Geary et al.

Summary: RNA origami is a framework for the modular design of nanoscaffolds that can be folded from a single strand of RNA and used to organize molecular components with nanoscale precision. The newly developed computer-aided design software has been shown to improve the folding yield of kilobase-sized RNA origami, simplifying the construction of custom RNA scaffolds for nanomedicine and synthetic biology. The design of genetically expressible RNA origami requires modeling and design tools that simultaneously consider thermodynamics, the folding pathway, sequence constraints and pseudoknot optimization.
