Gut Microbiota May Play a Significant Role in the Pathogenesis of Graves' Disease

Summary: This study identified differences in the composition and metabolism of gut microbiota in patients with autoimmune thyroid diseases, particularly Graves' disease. Lactobacillus may play a key role in the pathogenesis, and certain microbial genera showed significant correlations with thyroid function tests. The findings suggest that microbiota could be a potential noninvasive diagnostic tool for Graves' disease, and further research is needed to explore their role in autoimmune thyroid diseases.

Associations between gut microbiota and thyroidal function status in Chinese patients with Graves' disease

Summary: The study found that the gut microbiota diversity in GD patients was significantly lower than in healthy volunteers, with differences mainly at the genus level. With improvement in thyroid function, specific genus abundance also changed significantly. Spearman correlation analysis revealed positive and negative correlations between TRAb levels and certain bacterial genera.

Summary: The study compared T cell profiles between patients with persistent Graves' disease (pGD) and newly diagnosed GD (nGD), finding lower proportions of Th17 and Treg cells in pGD, along with decreased TCR diversity. pGD patients showed marked clonal expansion and more public T cell clonotypes. Additionally, upregulation of inflammation and chemotaxis pathways, coupled with downregulation of Th17 and Treg cell-associated genes, was observed in pGD.

Functional experiments indicated that carriers of the VDR/Apa I AA genotype had decreased VDR expression, reduced TGF-beta 1 production, and increased IL-17 production.

Predisposition to Graves' disease and Graves' ophthalmopathy by genetic variants of IL2RA

Summary: This study revealed the association between IL2RA/rs2104286 locus and the susceptibility to Graves' disease and Graves' ophthalmopathy, potentially affecting the occurrence of these diseases by influencing IL2RA gene mRNA expression and cytokine production. Other loci polymorphisms such as IL2RA (rs3118470, rs7093069), miR27a/rs895819, miR182/rs76481776, and FoxO1 (rs2297626, rs17592236, rs9549241, rs12585277) were not found to be associated with GD susceptibility.
