Immune signature-based risk stratification and prediction of immune checkpoint inhibitor's efficacy for lung adenocarcinoma

Ming Yi et al.

Summary: This study established an immune signature for lung adenocarcinoma (LUAD) patients, showing that patients with low-risk scores had better overall survival, differences in tumor-infiltrating lymphocytes and tumor mutation burden between high and low-risk groups, and a potential predictive relevance with immune checkpoint inhibitor (ICI) therapy.

Initiation of Pancreatic Cancer: The Interplay of Hyperglycemia and Macrophages Promotes the Acquisition of Malignancy-Associated Properties in Pancreatic Ductal Epithelial Cells

Lilli Otto et al.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive solid malignancy with poor prognosis, commonly associated with obesity and type 2 diabetes mellitus (T2DM), both characterized by high blood glucose levels. Macrophages play a significant role in PDAC development as the main immune cell population. This study reveals the impact of T2DM-related hyperglycemic, inflammatory microenvironment on the acquisition of malignancy-associated alterations in premalignant pancreatic ductal epithelial cells (PDEC), offering new insights into how metabolic diseases may promote PDAC initiation.

Tumor mutational burden assessment in non-small-cell lung cancer samples: results from the TMB2 harmonization project comparing three NGS panels

Javier Ramos-Paradas et al.

Summary: Tumor mutational burden (TMB) is a proposed predictive biomarker for immunotherapy in solid tumors. Two next generation sequencing panels, TSO500 and OTML, showed strong agreement with a reference assay FO in NSCLC samples. TSO500 exhibited higher inter-laboratory reproducibility and adjusted cut-off values ensured a sensitivity >88%.

Identification of VWF as a Novel Biomarker in Lung Adenocarcinoma by Comprehensive Analysis

Yi He et al.

Summary: In this study, differential co-expressed genes between lung adenocarcinoma (LUAD) and normal samples were explored using the Gene Expression Omnibus (GEO) dataset GSE43458 and The Cancer Genome Atlas (TCGA). A total of 366 co-expressed genes were identified, with VWF identified as a potential novel biomarker for prognosis of LUAD based on expression analysis and survival data.

A Novel Immune-Related Prognostic Model for Response to Immunotherapy and Survival in Patients With Lung Adenocarcinoma

Yujia Zheng et al.

Summary: This study focused on analyzing the immune infiltration status of lung adenocarcinoma (LUAD) and identified that the infiltration of M0 macrophage cells and follicular helper T cells predicts an unfavorable overall survival of patients. Through clustering based on immune scores, LUAD was classified into hot and cold tumors, each with distinct transcription profiles and enriched pathways. A Cox regression model was developed based on hub genes and prognostic-related genes to predict patient survival, showing potential for facilitating the clinical application of immunotherapy in LUAD.

Overexpression of PLXDC2 in Stromal Cell-Associated M2 Macrophages Is Related to EMT and the Progression of Gastric Cancer

Yiming Guan et al.

Summary: PLXDC2 is a gene associated with the tumor microenvironment, linked to poor prognosis and advanced disease in gastric cancer patients, and correlated with the expression of M2 macrophages and inflammatory signaling pathways. Its expression in stromal cells and interaction with tumor-associated macrophages may induce the EMT process, contributing significantly to cancer biology.

Tumor Cell-Derived Exosomal miR-770 Inhibits M2 Macrophage Polarization via Targeting MAP3K1 to Inhibit the Invasion of Non-small Cell Lung Cancer Cells

Jixian Liu et al.

Summary: The study identified that tumor cell-derived exosomal miR-770 inhibited M2 macrophage polarization via targeting MAP3K1, thus suppressing the invasion and growth of NSCLC cells. This provides a new strategy for the treatment of NSCLC.
