Summary: The study utilized a CKD/ApoE(-/-) mouse model to reveal that phenotypic switching and premature senescence of VSMCs are significant factors contributing to plaque vulnerability in CKD. Through multi-omics analysis and gene knockout experiments, it was confirmed that CKD-induced oxidative stress leads to mitochondrial damage in VSMCs, triggering an IFN-I response and resulting in vulnerable plaques. Blocking the IFN-I response might hold promise for treating cardiovascular diseases associated with CKD.
