Summary: This study reveals the detrimental role of CD8(+) T lymphocytes following acute myocardial infarction, showing that they release Granzyme B in the ischemic heart tissue leading to cardiomyocyte apoptosis, adverse ventricular remodeling, and deterioration of myocardial function. Depletion of CD8(+) T lymphocytes leads to reduced apoptosis, inhibited inflammatory response, limited myocardial injury, and improved heart function. Elevated circulating levels of GRANZYME B in AMI patients may predict increased risk of death at 1-year follow-up. Targeting pathogenic CD8(+) T lymphocytes could be a potential therapeutic strategy in the setting of acute myocardial infarction.

β1-adrenergic receptor N-terminal cleavage by ADAM17; the mechanism for redox-dependent downregulation of cardiomyocyte β1-adrenergic receptors

Summary: The study revealed that beta(1)-adrenergic receptors are expressed in cardiomyocytes and other cell types as both full-length and N-terminally truncated species, with the truncated species being formed as a result of O-glycan regulated N-terminal cleavage by ADAM17. It was also found that oxidative stress leads to redox-inactivation of cardiomyocyte beta(1)ARs through N-terminal truncation by ADAM17, and overexpression of a cleavage resistant beta(1)AR mutant exacerbates cell apoptosis. These findings suggest that the beta(1)AR N-terminus plays a crucial role in beta(1)AR responses and may be targeted for therapeutic advantage.

Alleviation of Inflammation and Oxidative Stress in Pressure Overload-Induced Cardiac Remodeling and Heart Failure via IL-6/STAT3 Inhibition by Raloxifene

Summary: In this study, inflammation and IL-6/STAT3 signaling were found to be activated in pressure overload-induced heart failure (HF) in mice. Prolonged IL-6 stimulation also induced oxidative stress and an increase in mitophagy-related proteins in H9c2 myoblasts, all of which were inhibited by raloxifene. These findings suggest that IL-6/STAT3 signaling may play a role in the pathogenesis of myocardial hypertrophy and HF.
