PRODIGE 59-DURIGAST trial: A randomised phase II study evaluating FOLFIRI plus Durvalumab ± Tremelimumab in second-line of patients with advanced gastric cancer

Camille Evrard et al.

Summary: The poor overall survival of patients with gastric or gastro-oesophageal junction adenocarcinomas has prompted research into first-line and second-line chemotherapy regimens, as well as the combination of immune checkpoint inhibitors with chemotherapy. Results have been mixed, highlighting the ongoing challenge in treating these types of tumors.

Phase III Trial of Avelumab Maintenance After First-Line Induction Chemotherapy Versus Continuation of Chemotherapy in Patients With Gastric Cancers: Results From JAVELIN Gastric 100

Markus Moehler et al.

Summary: The use of avelumab as maintenance therapy after first-line chemotherapy did not show superior overall survival compared to continued chemotherapy in patients with advanced GC or GEJC, regardless of PD-L1 status. Treatment-related adverse events occurred in both groups, with more severe events in the continued chemotherapy group.

Granulocyte-Macrophage Colony-Stimulating Factor-Activated Neutrophils Express B7-H4 That Correlates with Gastric Cancer Progression and Poor Patient Survival

Zhi-guo Shan et al.

Summary: The study revealed that neutrophil levels in tumor tissues of gastric cancer patients were significantly higher than in non-tumor tissues, positively associated with tumor progression but negatively correlated with patient survival. Most intratumoral neutrophils exhibited an activated phenotype and expressed the immunosuppressive molecule B7-H4. The GM-CSF-B7-H4 pathway plays a regulating role in gastric cancer.

Expression, regulation and clinical significance of B7-H3 on neutrophils in human gastric cancer

Zheng-Yan Li et al.

Summary: In gastric cancer tumors, neutrophils expressing CD54 and B7-H3 are closely associated with changes in GM-CSF levels, activating through the JAK-STAT3 signaling pathway. B7-H3+ neutrophils may potentially serve as biomarkers in gastric cancer, increasing with tumor progression and predicting overall survival.
