Schizophrenia-Linked Protein tSNARE1 Regulates Endosomal Trafficking in Cortical Neurons

Melissa Plooster et al.

Summary: TSNARE1, a high-confidence gene candidate for schizophrenia risk, regulates endosomal trafficking in cortical neurons and may act as an inhibitory SNARE by competing for incorporation into endosomal SNARE complex. Its localization in endosomal network and late endosomes suggests a role in negative regulation of early endosomal to late endosomal trafficking.

Heterozygous variants in SPTBN1 cause intellectual disability and autism

Jill A. Rosenfeld et al.

Summary: Spectrins are important components of the cytoskeleton, involved in protein localization, signal transduction, and cellular scaffolding. Variants in SPTBN1 have been associated with developmental delays, intellectual disability, and behavioral disturbances, expanding our understanding of human spectrinopathy disorders and highlighting the essential roles of SPTBN1 in neurodevelopment.

Missense and truncating variants in CHD5 in a dominant neurodevelopmental disorder with intellectual disability, behavioral disturbances, and epilepsy

Ilaria Parenti et al.

Summary: The CHD5 gene, located in the critical 1p36 microdeletion region, encodes a subunit of the NuRD complex required for neuronal development. Variants in this gene were identified in 16 unrelated individuals, showing features like intellectual disability, speech delay, and epilepsy.

Pathogenic SPTBN1 variants cause an autosomal dominant neurodevelopmental syndrome

Margot A. Cousin et al.

Summary: SPTBN1 mutations cause a neurodevelopmental syndrome with intellectual disability, language and motor delays, and other features. Heterozygous SPTBN1 variants disrupt beta II-spectrin stability, binding to molecular partners, and disturb cytoskeleton organization and dynamics, suggesting compromise of neural development and function. This study expands the understanding of spectrinopathies affecting the brain and highlights the critical role of beta II-spectrin in the central nervous system.
