Protectors of the Mitochondrial Permeability Transition Pore Activated by Iron and Doxorubicin

Tatiana A. Fedotcheva et al.

Summary: The study aimed to investigate the effects of iron, DOX, and their complex on MPTP opening and to find potential protectors for MPTP. The results identified MPTP and respiratory complex II as the main targets of DOX on isolated mitochondria, and various MPTP protectors were found to attenuate the impact of iron and its complex with DOX on MPTP opening, offering new strategies to mitigate the adverse effects of DOX on mitochondrial function.

Protective effects of three structurally similar polyphenolic compounds against oxidative damage and their binding properties to human serum albumin

Chuanying Zhang et al.

Summary: Brazilin, hematoxylin, and hematein are polyphenols with similar structures and rich biological activities, and they have protective effects against oxidative degradation of human serum albumin. The biological effects of these polyphenols are mainly influenced by their binding to protein, and fluorescence experiments indicate that they bind to protein through a static quenching mechanism, causing some conformational changes. These findings provide insights into the role of molecular structure in the protective effects and binding abilities of polyphenols against protein oxidative damage.

The Multifaceted Regulation of Mitochondria in Ferroptosis

Hao Wu et al.

Summary: Ferroptosis is a novel form of regulated cell death initiated by lethal lipid peroxidation catalyzed by cellular labile free iron. It differs significantly from other regulated cell death forms at morphological, genetic, and biochemical levels. Dysregulated ferroptosis is highly relevant to the pathogenesis of degenerative diseases and organ injuries in human patients, and the targeted induction of ferroptosis is considered a potential therapeutic strategy for therapy-resistant cancers. Mitochondria play a crucial role in regulating ferroptosis, with compromised mitochondrial morphology and functionalities primarily involved in this process. Various mitochondria-associated proteins and metabolic processes have been identified to modulate ferroptotic cell death.

Influence of Microbial Metabolites on the Nonspecific Permeability of Mitochondrial Membranes under Conditions of Acidosis and Loading with Calcium and Iron Ions

Nadezhda Fedotcheva et al.

Summary: Mitochondrial dysfunction is seen as a major factor in multiple organ failure caused by chronic inflammation and sepsis. Microbial metabolites have been found to affect the bioenergetic processes in mitochondria, influencing the permeability of mitochondrial membranes under pathological conditions. These metabolites can directly impact the nonspecific permeability of mitochondrial membranes when conditions of acidosis, an imbalance of calcium ions, and excess free iron are present.
